Hello,
I've been trying to set up the tools and zeros for my lathe with centroid cnc 12 and the spindle will not turn when I hit the turn buttons. It will jog and seems to do everything else.

You have to put the spindle into manual mode before the spindle start/stop buttons will work. Second button in the top row of the VCP is labeled "Auto Spindle Man". This switches the spindle between auto and manual modes. See page 20 in the lathe operator's manual.
