Enclosure size can be determined by laying out all your components on the bench. As for connectors and such, that depends on what you are connecting up. Your question on what to use is pretty broad and application specific.
Look at pictures and videos of others CNC control cabinets for ideas. Go check out the Centroid success stories forums, maybe there are some ideas there

You need a motor power supply to drive the Clearpath motors.
I have not wired up the system yet, as I am still waiting for the ClearPath system. I will use their (Teknic's) power supply, and will use the Centroid PS to power the Acorn board.

That said, Marty can you tell me what relay block are you using your control box shown in the YT video Emco PC5 Centroid CNC Acorn Conversion: Part 7 Cabinet done. Overview?

What motor voltage are you using and AC or DC. That build was a 24vdc 10A axis motor power supply.

I am using the ClearPath SDSK series with their IPC-5 at 75 volts.

What kind of current?
Point being is you have to match the contactor to the type of voltage and the load.
You have to make sure the contactor is rated for DC voltage and the current running through it. If you get a 3 pole contactor you can run the DC positive and DC negative through it across two contacts spreading the load. The third pole can be used to break the VFD common as shown in the Acorn system schematic.

Look for a new MOELLER EATON DILM32-01 on eBay.
It uses a 24vdc coil and should be more than enough.
You should use separate power supply for it. The logic power supply included with Acorn is only large enough for Acorn and perhaps some logic circuitry.

Always use snubbers across relay coils.

The NEMA34 on the z axis is 273W, and will draw 3 A and 75V, while the two NEMA23 on x and y will be 173W, drawing 2.3 A each, also at 75V. The spindle is 1HP so on 120 V should draw about 6-7 Amps.

So when the e-Stop is activated, I send the signal to the Acorn from one side of the e-Stop and to the contactor from teh other side of the e-Stop. Will the contactor turn off the spindle only or should I also stop the x-, y-, and z- axis motors?

What is a snubber across the relay poles (Sorry for ignorance)?
